Resolve which strand was written down
A coding strand and a template strand describe different relationships to RNA. Replacing thymine with uracil is appropriate for a coding strand written in the matching direction. A template requires complementation, and its written orientation determines whether reversal is needed first.

A coding strand becomes RNA by replacing T with U. A template strand is complemented; a 5′→3′ template is reversed before complementation.
Retain the input definitions and units with the result. A later comparison should use the same basis; a change in method is a new scenario, not simply a more precise version of the old number.
